凱氏定氮原理分析
KDN係列定氮儀是浙江托普基於凱式定氮法而自主研發的新一代全自動凱式定氮儀。凱(kai)氏(shi)定(ding)氮(dan)法(fa)是(shi)基(ji)於(yu)分(fen)析(xi)化(hua)學(xue)的(de)滴(di)定(ding)分(fen)析(xi)方(fang)法(fa)得(de)出(chu)的(de)。在(zai)測(ce)定(ding)物(wu)質(zhi)中(zhong)氮(dan)元(yuan)素(su)含(han)量(liang)時(shi),由(you)於(yu)被(bei)測(ce)物(wu)質(zhi)不(bu)能(neng)直(zhi)接(jie)與(yu)滴(di)定(ding)劑(ji)發(fa)生(sheng)化(hua)學(xue)反(fan)應(ying),所(suo)以(yi)通(tong)過(guo)其(qi)它(ta)反(fan)應(ying)以(yi)間(jian)接(jie)的(de)方(fang)式(shi)測(ce)定(ding)被(bei)測(ce)物(wu)質(zhi)中(zhong)氮(dan)的(de)含(han)量(liang)。
根據全自動定氮儀凱氏定氮法,在測定氮元素含量時分成如下3個步驟:
(1)消煮:測ce定ding物wu質zhi中zhong的de氮dan元yuan素su含han量liang時shi,通tong常chang將jiang試shi樣yang做zuo適shi當dang處chu理li,將jiang各ge種zhong氮dan化hua合he物wu分fen解jie並bing轉zhuan化hua為wei簡jian單dan的de銨an根gen離li子zi。采cai用yong的de方fang法fa為wei取qu一yi定ding的de試shi樣yang,置zhi於yu消xiao煮zhu管guan中zhong,與yu濃nong硫liu酸suan及ji催cui化hua劑ji(硫酸銅、硫酸鉀)在高溫下充分煮解,生成硫酸銨。
(2)蒸餾:在(zai)消(xiao)煮(zhu)後(hou)完(wan)全(quan)分(fen)解(jie)的(de)含(han)銨(an)根(gen)離(li)子(zi)的(de)試(shi)樣(yang)中(zhong)加(jia)過(guo)量(liang)的(de)濃(nong)氫(qing)氧(yang)化(hua)鈉(na)溶(rong)液(ye)加(jia)熱(re)蒸(zheng)餾(liu),使(shi)銨(an)根(gen)離(li)子(zi)轉(zhuan)化(hua)為(wei)氨(an)氣(qi)。反(fan)應(ying)中(zhong)釋(shi)放(fang)的(de)氨(an)氣(qi)與(yu)水(shui)蒸(zheng)汽(qi)一(yi)起(qi)經(jing)冷(leng)凝(ning)管(guan)冷(leng)凝(ning),被(bei)收(shou)集(ji)在(zai)接(jie)收(shou)杯(bei)中(zhong)的(de)硼(peng)酸(suan)混(hun)合(he)吸(xi)收(shou)液(ye)中(zhong),蒸(zheng)餾(liu)出(chu)來(lai)的(de)氨(an)氣(qi)可(ke)用(yong)過(guo)量(liang)但(dan)不(bu)需(xu)計(ji)量(liang)的(de)硼(peng)酸(suan)溶(rong)液(ye)吸(xi)收(shou),形(xing)成(cheng)蒸(zheng)餾(liu)接(jie)收(shou)液(ye)。
(3)滴定:用鹽酸標準滴定溶液滴定生成的硼酸二氫銨溶液,選用溴甲酚綠與甲基紅混合溶液作為指示劑。
消煮與蒸餾2個ge階jie段duan的de實shi質zhi就jiu是shi將jiang不bu能neng與yu滴di定ding劑ji鹽yan酸suan直zhi接jie反fan應ying的de被bei測ce物wu質zhi轉zhuan換huan為wei可ke以yi與yu鹽yan酸suan反fan應ying的de化hua合he物wu,並bing且qie該gai化hua合he物wu中zhong氮dan的de質zhi量liang就jiu是shi被bei測ce物wu質zhi中zhong氮dan的de總zong質zhi量liang。第di3階(jie)段(duan)就(jiu)是(shi)用(yong)鹽(yan)酸(suan)標(biao)準(zhun)滴(di)定(ding)溶(rong)液(ye)滴(di)定(ding)生(sheng)成(cheng)的(de)硼(peng)酸(suan)二(er)氫(qing)銨(an)溶(rong)液(ye),但(dan)鹽(yan)酸(suan)與(yu)硼(peng)酸(suan)二(er)氫(qing)銨(an)溶(rong)液(ye)恰(qia)好(hao)完(wan)全(quan)反(fan)應(ying)時(shi)通(tong)常(chang)沒(mei)有(you)明(ming)顯(xian)的(de)外(wai)部(bu)特(te)征(zheng),一(yi)般(ban)是(shi)根(gen)據(ju)指(zhi)示(shi)劑(ji)的(de)變(bian)色(se)來(lai)確(que)定(ding)滴(di)定(ding)終(zhong)點(dian)的(de)。另(ling)外(wai),根(gen)據(ju)各(ge)種(zhong)蛋(dan)白(bai)質(zhi)中(zhong)皆(jie)各(ge)有(you)固(gu)定(ding)比(bi)數(shu)的(de)氮(dan)的(de)事(shi)實(shi),求(qiu)出(chu)氮(dan)的(de)含(han)量(liang),乘(cheng)以(yi)相(xiang)應(ying)的(de)換(huan)算(suan)因(yin)子(zi),就(jiu)是(shi)粗(cu)蛋(dan)白(bai)的(de)含(han)量(liang)。這(zhe)也(ye)是(shi)蛋白質測定儀(粗蛋白測定儀) 測量物體蛋白質含量的根本原理。
